Network for Therapeutic Nucleic Acids
Therapeutic nucleic acids or oligonucleotides have enormous potential for the treatment of currently untreatable diseases, for the targeted and gentle treatment of serious diseases and for patient-specific therapy.
However, the biggest hurdle is the step from the laboratory to the clinic, as complementary issues need to be addressed and other skills and experience are required.
Mission
Transfer of patient-specific therapies from the development to the clinic:
- Realising the treatment of currently untreatable diseases, for targeted and mild treatment of severe diseases and patient-individualized therapy
- Bringing together developers for therapeutic nucleic acids, developers of novel formulations and devices, manufacturers, analysts and clinicians
- Processing of questions regarding acceptance criteria for GMP material, validation parameters for analytical methods, formulations or inclusion criteria for clinical trials
- Connecting developers of new therapeutic ideas with the production of clinical trial material and clinical trial centres
- Facilitating entry into clinical trials
Development needs
The network focuses on the following development lines:
- Sequence optimisation of therapeutic nucleic acids with regard to their stability, purity and efficient producibility while maintaining high efficacy
- Chemical synthesis and analysis of long oligonucleotides (<100 bases)
- Formulation, controlled release and targeted administration of therapeutic nucleic acids
- Analysis of oligonucleotides in therapy and diagnostics
- Environmentally friendly, sustainable and scalable production of therapeutic nucleic acids
- Planning, implementation and evaluation of clinical studies for therapeutic nucleic acids and development of a study database to optimise follow-up studies
- Combinations of therapeutic nucleic acids and other active ingredients

- 1. Delivery and FormulationNovel non-viral lipid- and polymer-based vectors, oral administration, sustainable manufacturing concepts, alternatives to critical excipients
- 2. Nucleotide DevelopmentTherapeutic oligonucleotides including silencing oligonucleotides, mRNA and novel formulations
- 3. Analysis and PurificationDevices and services, HPLCs with specific detectors and column combinations, developments in the field of cross-flow filtration
- 4. ProductionDevices and services, scalable synthesizers, optimised HPLC systems, large-scale ultrafast cross-flow filtration, recycling systems, formulation optimisation, solvent alternatives in production
- 5. Preclinical DevelopmentOptimised sequences, modifications and delivery – proof-of-concept/mode-of-action studies
- 6. Clinical DevelopmentSoftware/AI-based evaluation of completed studies (data mining), projection of the results obtained for the planning of future studies, support in the planning of studies for therapeutic oligonucleotides including software-based risk assessment
Network partners
In addition to developers of therapeutic nucleic acids and oligonucleotides, developers of novel formulations, device developers for the production and analysis of oligonucleotide active ingredients and drugs, manufacturers of oligonucleotides and formulations as well as clinics are also to be networked.
